This quest is initiated by examining a Black Book on a Table at +1.48, -12.75, -102.48 in the Temple of Cazic-Thule. This is the room with the Ancient Cadavers.

You must slay 20 Spinechiller Broodtenders in the Temple of Cazic-Thule. These are the spiders in the courtyard/temples area of the zone.

This quest is specific. Not all the spiders in that area count toward the update. Just an FYI.
